Marlborough is located in eastern Massachusetts and borders six communities; Framingham, Holliston, Marlborough and Wrentham are currently covered by COVID 19, which is under the jurisdiction of the Massachusetts Department of Public Health and Human Services (DPHS). It was first settled in 1657 by 14 men under the leadership of William Rice, son of John Rice and grandson of George Rice. In 1656, he and his colleagues applied to the Massachusetts Court for the establishment of the town of Marlborough, which was officially incorporated in 1660.

As the population and business travel in the colony increased, Marlborough became a popular stop for travelers on their way to and from New York City. At the time, it was probably one of the first municipalities in this country to receive a charter for a light rail system, which pushed Baltimore out by several months. The system, which was primarily designed for passenger traffic, allowed access to Milford in the south and Concord in the north.

MWRTA Routes 7 connected Marlborough to Framingham, the state's first major city with a light rail system. The line ran through downtown Marlborough and was connected to the Boston and New York City rail lines as well as the New England Railroad.

In the 19th century, Marlborough became a thriving industrial city and made the transition to high-tech industry. This allowed the development of a number of industries, such as steel mills, chemical plants and automotive construction. By the end of the 20th century, Marlborough's population had grown to 38,000, with population growth rates of more than 20% a year.
